Scientific work over the past 30 years revealed that
SOS protein is directly involved in the activation
of ras-p21.The aim of this research project was to
synthesize a plasmid which would ultimately express
a SOS peptide, 994-1004, which was proven to inhibit
the Val 12 ras-p21 pathway in Xenopus laevis
oocytes. The SOS plasmid consisted of a vector,
pOPRSVI/MCS, ligated to DNA oligonucleotides
corresponding to the SOS peptide of interest.
Different SOS plasmid concentrations, ranging from
1ug/uL to 5.138ug/uL, were either co-microinjected
with Val 12 p21 or microinjected into oocytes
followed by incubation in insulin or in
progesterone. At the 48th hour time point, the
results showed that the 3.225ug/uL SOS plasmid gave
the most inhibition on the oncogenic pathway while
the wild-type p21 pathway was unaffected by this
concentration. This data suggest that the 3.225
ug/uL SOS plasmid is a specific inhibitor of the
oncogenic ras-p21 pathway.
